Abstract: ABSTRACT: Melatonin (MT) is primarily associated with sleep regulation and plays crucial roles in maintaining bone health. This hormone emerges as a promising alternative in the treatment of osteoporosis (OP), especially in postmenopausal women. This study is an integrative literature review, encompassing works published in Portuguese or English from 2013 to 2023 in the PubMed, ScienceDirect, and Medline databases. It was conducted using descriptors such as Melatonin, Osteoporosis, Postmenopause, Menopause, and Ovariectomy, combined with the boolean operator AND. Out of a total of 993 identified publications, 12 were selected after applying inclusion and exclusion criteria. Among these, two were double-blind randomized studies with women, and the remaining ten involved rodents. The studies highlight the effectiveness of MT in preserving and improving bone health in animal models, emphasizing its ability to increase osteoblastic differentiation in vitro and promote positive results in bone density in rodents and women. MT supplementation in postmenopausal women revealed physical and psychological benefits, with few adverse effects and high user adherence. MT emerges as a potential supplement to preserve bone mass density in women at this stage and possibly in other life stages. However, the precise relationship between MT and bone homeostasis still lacks complete elucidation, requiring further studies to understand its mechanism of action, as well as its effectiveness in isolation or in combination with other substances such as calcium and vitamin D3.
