USO DA METFORMINA E DE PUNICA GRANATUM L. (ROMÃ) NO APARELHO REPRODUTOR DE RATAS WISTAR COM SÍNDROME DO OVÁRIO POLICÍSTICO
A Síndrome dos Ovário Policístico (SOP) é a principal endocrinopatia ginecológica na idade reprodutiva em mulheres. Muitos são os sintomas e variáveis para cada paciente. Sendo assim, o mesmo acontece com o tipo de tratamento, pois irá depender da gravidade da disfunção endócrina e do objetivo da paciente. Os tratamentos mais utilizados são o uso de contraceptivos orais combinados e os agentes insulino-sensibilizantes como a metformina que, embora apresente bons resultados, também exerce muitos efeitos colaterais. Assim, destacam-se inúmeros tratamentos alternativos, como o extrato de folha de romã, que objetivou o presente trabalho. Comparou-se dois tipos de tratamento para a SOP: um clássico e outro alternativo. Foram utilizados 20 ratos Rattus novergicus (linhagem Wistar), fêmeas, recém-nascidas, que, aos cinco dias de vida, entraram no processo de indução à SOP com dose única de 10 mg/Kg de Testosterona (Durateston) diluído em álcool benzílico (9,45mg/mL) via subcutânea. Após a confirmação da SOP por meio de lavagens vaginais e a permanência na fase de estro, os animais foram divididos em dois grupos de 10 animais cada, para o início dos tratamentos propostos com a metformina e o extrato de folha de romã por gavagem durante 21 dias. Ao final deste período os animais foram sacrificados de acordo com as normas do CEPan (Nº 418/20), feita a laparotomia e a retirada dos ovários para os procedimentos histopatológicos de rotina. Na análise histopatológica dos ovários observou-se a presença de folículo primordial, folículo atrésico, corpo lúteo e cistos em ambos os tipos de tratamentos, sem diferença significativa em nenhuma das fases evolutivas dos folículos, podendo inferir que, no tempo em que foi realizado os tratamentos, ambos apresentam eficácia semelhante.

Abstract:
The Polycystic Ovary Syndrome (PCOS) is the main gynecological endocrinopathology in reproductive age women. There are many symptoms and variables for each patient. Therefore, the same happens with the type of treament used because it will depend on the endocrine’s dysfunction’s severity and the patient’s goal. Combined oral contraceptives and insulin-sensitizers are the most used treatments that, although showing good results, also has many side effects. Thus, there are lots of alternative treatments, such as pomegranate´s leaf extract, which is presented in this study. Two types of PCOS’s treatments were compared: a classic and an alternative one. 20 newborn female rats Rattus novergicus (Wistar lineage) were used. At five days old, they entered at the PCOS’s inducing process by a single dosage of 10 mg/Kg Testosterone (Durateston), diluted in benzyl alcohol (9,45 mg/mL) subcutaneously. After the PCOS’s diagnose was confirmed by collection of vaginal fluid washings and being on estrus phase permanently, the animals were divided into two groups with 10 animals each. The proposed treatments begun using metformin and pomegranate’s leaf extract by gavage for 21 days. By the end of this period, the animals were sacrificed according to the rules of CEPan (Nº 418/20), laparotomized and the ovaries were removed for routine histopathological procedures. In the histopathological analysis of the ovaries, primordial follicles, atresic follicles, corpus luteum and cysts were found in both types of treatments, without a significant difference in any evolutionary phase of the follicle, concluding that, by the treatment’s period of time, both displayed similar efficiency.
