Abstract: Analysis of the composition of gallstones is vital for the etiopathogenesis of gallstone disease and can help prevent its formation. In this study, an in vivo gallstone sample was characterized ex vivo using solid-state 13C nuclear magnetic resonance (NMR) magic angle spinning (CPMAS) sequence in prospecting the majority constituents present, with minimal material manipulation. The acquisition conditions were optimized, and the sample preparation involved only its grinding in an agate mortar. It was found that the gallbladder stones in the present study had cholesterol as the main constituent. A comparison of 13C chemical shifts suggested that the predominantly polymorphic form would be crystalline cholesterol monohydrate, usually found in patients with chronic cholecystitis.
