Abstract: Agriculture faces the challenge of increasing food production for an exponentially growing population, even in the face of the challenges imposed by climate change. Water scarcity leads to fluctuations in water availability for crops, affecting their full development and consequent productivity. Science has sought to understand the mechanisms involved in drought tolerance in plants in order to develop increasingly tolerant genotypes. Along the same lines, the objective of this study was to use the CRISPR/Cas9 gene editing technique to obtain a genetic construct aimed at knocking out the OsDREB1C gene, a promising candidate for increasing drought tolerance in rice plants (Oryza sativa L.). To this end, specific spacers for OsDREB1C were designed and cloned into the pRGEB31 vector in the 5' portion of the gRNA backbone. The plasmid was multiplied in E. coli and purified for later confirmation by restriction assay with the enzymes BsaI and SacI and Sanger sequencing. Sequencing confirmed the cloning and integrity of the spacer, confirming an integral construct to be used in the transformation of rice plants. Thus, it is concluded that it was possible to obtain an integral construct for the knockout of the OsDREB1C gene. The analysis of plants containing this construct will allow studying the functions of this transcription factor in plants subjected to water stress.
