Abstract: Genetic mutations that occur randomly during DNA replication can lead to the formation of cancer cells. Chronic Myeloid Leukemia (CML) is an cancer caused by a mutation in the Philadelphia chromosome, which results in the excessive production of myeloid cells. Available treatments have a narrow therapeutic range and greater side effects, requiring the use of more effective alternatives. Tyrosine kinase inhibitors, such as imatinib, which inhibit the BCR-ABL protein, are promising for offering a better quality of life to patients. This review aims to compile and discuss the importance of therapies for CML, focusing on the evolution of available treatments. The methodology consisted of a bibliographic review using the SCIELO, ScienceDirect and Google Scholar databases, using as inclusion criteria those studies that addressed the characteristics of CML and its pharmacotherapy. The treatment of CML was revolutionized by the introduction of tyrosine kinase inhibitors, such as imatinib, which inhibit the BCR-ABL protein, offering patients a better quality of life. However, until the arrival of tyrosine kinase inhibitors, other drugs were used, such as busulfan, hydroxyurea and interferon alpha, which were less effective and had a higher mortality rate. The evolution in the treatment of CML represents a milestone in oncology, highlighting the need for targeted and personalized therapies. Therefore, it is essential to develop new generations of these drugs to overcome the resistance developed by cancer cells, improving treatment adherence and clinical outcomes for patients.
