Abstract: There are microorganisms with great infectious potential in all environments, today we already know that many of them make up the human microbiota, but they live in symbiosis or mutual relationship. On the other hand, the physiological condition of the host, when compromised, can lead to an imbalance in this relationship, and the same microorganisms can lead to a pathological condition. Therefore, through a practical laboratory project, this work aims to develop a biochemical test method for Gram positive bacteria of the genus Staphylococcus and Enterococcus in microplates. Using as a basis the tests of Urea, Lactose, Glucose, Maltose, Sarcrose and Fructose. The work had a relevant result due to the verification of positive results in the inoculation of bacterial strains inside the microplates, with a color change allowing a possible bacterial identification. Concluding that the use of this highly effective biochemical test method for Gram positive bacteria, in addition to reducing laboratory costs, also helps in practicality within the laboratory, seeing that a single plate can be used for several tests and for several patients.
