Abstract: Through the new molecular techniques a DNA extraction strategy was developed from cryopreserved bovine semen in order to determine the quality and usefulness of DNA. In this work the sperm material of 15 reproductively active bovine stallions was used, each stallion was subjected to the semen extraction process using an electroejaculator, this is responsible for stimulating the accessory glands of the reproductive system of the bovine male by waves of low electrical charge, each semen sample was collected in graduated tubes, a macroscopic analysis and a spermiogram of each sperm sample obtained from the different stallions was performed, the samples were diluted in equal parts with 3 different cryoprotectants, empajilló adjusted the sperm load to 20 million and The straws were frozen by slow cooling in nitrogen vapors. For the extraction process of frozen semen DNA, two protocols were used, the traditional method of phenol-chloroform with proteinase k (modified) and the commercial kit of TRIzolTM Reagent. The DNA obtained under the two protocols was used to measure its quality and purity with a NanoDrop® 2000 and a fragment of the 16S rDNA gene was amplified to confirm its usefulness in PCR amplification of the genes of interest.
