Abstract: Glyphosate is the active compound of several pesticides, and its use has been associated with concerns regarding metabolic health issues. Maternal exposure to pesticide compounds may induce changes in the development of the offspring. This study aimed to evaluate the morphology and morphometry of neuromuscular junctions (NMJs) in the extensor digitorum longus (EDL) muscle of the offspring of mice exposed to glyphosate during the prenatal and postnatal periods, and to investigate whether the effects of this exposure would be exacerbated by a high-fat diet. The animals were divided into four groups: Control Group (CTL-F1), High-Fat Diet Group (HIPER-F1), Glyphosate Group (GF-F1), and Glyphosate + High-Fat Diet Group (GF-HIPER-F1). At 150 days of age, the animals were weighed and euthanized; obesity was assessed using the Lee index, and the EDL muscle was collected and stored in Karnovsky's fixative, then subjected to the nonspecific esterase histochemical technique for the morphological and morphometric analysis of the NMJs. The HIPER-F1 group showed a 28% increase in body weight and a 455% increase in retroperitoneal and periepididymal fat compared to the CTL-F1 group. The GF-HIPER-F1 group showed a 35% reduction in retroperitoneal and periepididymal fat. The GF-F1 group showed a 32% increase in area and a 15% increase in major diameter when compared to the CTL-F1 group. The minor diameter showed no changes in any group. The other groups (HIPER-F1 and GF-HIPER-F1) did not show significant changes.
