Abstract: Ruta graveolens L., popularly known as rue, is a species widely used due to its medicinal, aromatic, and ornamental properties. Its bioactive compounds confer phytotherapeutic actions, increasing its economic potential and the need for studies on its ideal cultivation conditions. Thus, the present study aimed to verify the germination and germination speed index of rue (Ruta graveolens L.) in two substrates, in order to verify the best condition provided by the substrates for moisture retention and promotion of seedling growth and development. The experiment was conducted in greenhouses of the Fluminense Federal University (UFF), on the Gragoatá campus, Niterói/RJ, during December 2024. Two 180-cell trays were used, one for each treatment (substrates). The first substrate was Terra Vegetal from the Geolia® brand and the second was Terral Plantio from Terral Jardinagem®, which had an organic mineral compound - containing peat, vermiculite, perlite, manure, charcoal, thermophosphate, limestone and carbonized rice husk - maintained under controlled conditions. Germination began first in the substrate containing Terra Vegetal and showed a practically zero germination rate in the first days of counting in Terral Plantio, but both stabilized on the 20th day after sowing. The seeds cultivated in the organic mineral substrate (Terral Plantio) showed late germination, with a low germination rate and seedling development. The substrate containing only Terra Vegetal proved to be more suitable for the germination and initial growth of the rue seedling, providing greater seedling survival.
