Abstract: Objective: To compare the efficacy and safety of the CYD-TDV (Dengvaxia®), TAK-003, and Butantan-DV vaccines, highlighting their characteristics, mechanisms of action, immunological responses, and limitations, based on clinical trials published between 2017 and 2024. Methods: A narrative literature review was conducted in the PubMed (National Library of Medicine) database. The descriptors “dengue,” “vaccine,” and “treatment” were used, combined by the Boolean operator AND. Articles in English and Portuguese, published between 2017 and 2024, classified as clinical trials, randomized controlled clinical studies, or journal articles, were included. After screening, 21 articles met the inclusion criteria and were analyzed for vaccine efficacy, safety, and applicability. Results: CYD-TDV demonstrated an overall efficacy of 67.5%, with greater protection in previously exposed individuals but an increased risk in seronegative individuals. TAK-003 showed an average efficacy of 80%, uniform protection across serotypes, and less reliance on prior screening. Butantan- DV, based on inactivated virus, demonstrated robust immunogenicity and satisfactory safety, but requires further large-scale evidence. Conclusion: TAK-003 and Butantan-DV vaccines exhibit greater potential for widespread use due to their superior efficacy and safety, while CYD-TDV requires prior serological screening. Expanding vaccination and improving diagnostics are crucial for sustainable dengue control.
