Abstract: Introduction: The buccal mucosa is a very promising site of action for the administration of active substances and, therefore, has become very attractive as a target for formulations for local and systematic drug delivery. This route of administration has some advantages when compared to the oral route, it is possible to avoid gastric metabolism, the enzymatic manipulation of the drug and, mainly, to avoid first-pass metabolism. Objective: Discuss oral occupation as an alternative route of administration and to present its main conventional and nanostructured pharmaceutical forms. Methodology: This is a literature review. The databases used were: ScienceDirect, MDPI, Springer and Pubmed in the last 10 years of publication. Articles published in full in English and Portuguese were included. Inclusion criteria were articles published in full in English, Portuguese and Spanish. Incomplete publications, duplicated between databases, were excluded. Results: There are many pharmaceutical forms for oral administration in a conventional way, such as tablets and films. The discovered nanoparticles were: polymeric nanoparticles, nanostructured lipid carriers (NLC), solid lipid nanoparticles, PLGA nanoparticles coated with chitosan, metallic nanoparticles (gold, silver and palladium). Conclusion: The oral cavity is considered an excellent route of administration due to the ease of absorption of drugs by the oral mucosa. There are several conventional and nanostructured pharmaceutical forms for application in the oral mucosa.
